Background:

This test build is designed to utilize fluorescent in situ hybridization (FISH) testing methodology on blood lymphocytes for targeted conditions, such as aneuploidy.  Once the FISH data has been completed and reviewed, the test build is designed to immediately reflex to the most appropriate confirmatory or follow-up test.  Typically, if an abnormality is detected by FISH, then the reflex will be for high resolution chromosome analysis to confirm the abnormal FISH result.  If the FISH yields a normal result, then the reflex will be for chromosomal microarray testing for a more thorough genome-wide analysis to detect copy number changes and/or loss of heterozygosity, which may not be visible by high resolution analysis.

Methodology:

Slides are prepared per standard protocols and 100 interphase cells are scored for each probe under fluorescence microscopy.

Specimen Requirements:

Blood:  An EDTA AND a NaHep sample are both required for testing.
NaHep
  • Adult: 3-5 mL drawn into a GREEN top sodium heparin vacutainer tube or into a pre heparinized plastic syringe (use 0.2 cc sodium heparin, 1000 unit/mL). Do NOT use lithium heparin.
  • Child - 1-2 mL, as above.
  • Infant - 1-2 mL, as above. 
EDTA
  • Adult: 3-5 mL drawn into a PURPLE top EDTA vacutainer tube.   
  • Child: 1-2 mL, as above.
  • Infant: 1-2 mL, as above

Turn Around Time:

12-20 days. Preliminary FISH results will be issued prior to reflex testing. Final results will be reported after completion of FISH and reflex testing.

Shipment Sensitivity Requirements:

  • Keep specimen at room temperature during transit. 
  • Do not use the cold pack provided in the KDL shipping kit. 
  • Ship the specimen overnight express, using the FedEx priority overnight label provided. 
  • The specimen must arrive at the lab no more than 24 hours after collection.
